What this marker is

Proline is the amino acid that makes collagen possible. Collagen's triple helix depends on proline and its modified form hydroxyproline appearing at regular intervals along the chain, and since collagen is roughly a third of all protein in the body, that makes proline structurally central.

The modification step matters as much as the supply. Converting proline to hydroxyproline requires vitamin C, and without it the collagen produced is unstable. That single dependency is the mechanism behind scurvy.

Proline is not essential in the dietary sense, since the body synthesises it from glutamate. Demand during tissue repair can nonetheless outrun what synthesis provides.

If your result is low

Below 78 umol/L indicates low proline. Because collagen is the main consumer, the effects relate to connective tissue: slower wound healing, joint discomfort and skin quality.

Demand rises during recovery from injury or surgery, during intensive training that stresses tendons and ligaments, and with age as collagen synthesis becomes less efficient.

Modern diets contribute by omitting the collagen-rich cuts and preparations that traditional cooking included, so intake falls at the same time as the requirement stays constant.

If your result is in range

Between 78 and 550 umol/L your proline sits inside the reference range for this panel.

The range is wide because proline moves with recent intake, particularly of collagen or gelatin. Read it alongside glutamine, since the ratio between them describes the balance between gut repair and connective tissue repair demands.

If your result is high

Above 550 umol/L usually reflects recent collagen or gelatin intake, which raises proline substantially and quickly.

A high result is not concerning in itself. Proline is well tolerated and the body regulates it.

Where a high result appears without collagen intake, it is worth reading alongside liver function, since the liver handles amino acid metabolism. Raise that with your practitioner.

Why the Western lifestyle moves it

Western cooking moved away from the whole-animal preparations that supplied proline. Bone broth, slow-cooked joints and gelatin-rich cuts were routine and are now occasional, while muscle meat became the default.

That shift matters because muscle meat is comparatively low in proline and glycine. Eating more protein does not by itself restore what the collagen-rich preparations provided.

What to eat

Bone broth is the richest whole-food source and the most direct way to raise intake.

Slow-cooked meats on the bone, skin-on poultry and gelatin-rich preparations all supply proline in usable quantities.

Eat vitamin C-rich food alongside, since the conversion to hydroxyproline cannot happen without it. Bell peppers, citrus, kiwi and berries all work.

Supplement forms and doses

Collagen peptides at 10 to 20 g daily supply proline, hydroxyproline and glycine together in a hydrolysed and readily absorbed form.

Type I and III collagen are the forms associated with skin, tendon and bone. Type II is the one used for joint cartilage specifically.

Vitamin C at 500 to 1,000 mg daily is a requirement rather than an optional addition, because collagen synthesis stalls without it. Hyaluronic acid at 120 to 200 mg is sometimes added for joint and skin support.
