What this marker is

Glutamine is the preferred fuel of the cells lining the gut. Proline is the amino acid collagen is built from. This ratio compares the two, which puts two different repair demands on the same scale.

Both are conditionally essential: the body normally makes enough, but not during illness, injury, heavy training or sustained gut inflammation. That is exactly when the balance between them starts to say something.

On this panel the in-range band runs from 1.1 to 4.8, with 2.0 to 3.6 treated as optimal.

If your result is low

Below 1.1 means glutamine is low relative to proline, which points at gut demand drawing the glutamine pool down.

Prolonged intense training, illness, and anything irritating the intestinal lining all raise glutamine consumption. Because muscle is the main glutamine store, a sustained shortfall is often accompanied by muscle breakdown as the body mobilises it.

Read it alongside your individual glutamine result. A low ratio with low glutamine is a supply problem; a low ratio with normal glutamine is more about proline being high.

If your result is in range

Between 2.0 and 3.6 the balance sits in the optimal band, with 1.1 to 4.8 reading as in range.

Bone broth is the food that supplies both in roughly the proportion this ratio wants, which is why it appears in the recommendations for either direction.

If your result is high

Above 4.8 means proline is low relative to glutamine. Connective tissue repair is the side under-supplied.

Modern diets contribute directly here. Muscle meat is comparatively low in proline and glycine, and the collagen-rich cuts and preparations that traditional cooking used have largely dropped out.

Vitamin C matters as much as proline itself, because the conversion to hydroxyproline that stabilises collagen cannot happen without it. A high ratio alongside poor wound healing is worth checking vitamin C intake against.

Why the Western lifestyle moves it

Both sides are affected by the same pattern from different directions. Processed food, alcohol and frequent NSAID use raise the repair burden on the gut lining, which draws glutamine down.

Meanwhile the shift from whole-animal cooking to muscle meat cut proline intake without anything replacing it, so the other side of the ratio fell too.

What to eat

Bone broth supplies both in balance and is the simplest single change for either direction.

For the glutamine side: grass-fed beef, chicken, fish, eggs, dairy, cabbage and beets, plus fermented foods for the gut ecology that determines how much repair is needed.

For the proline side: slow-cooked meats, skin-on poultry and gelatin-rich preparations, with vitamin C-rich food in the same meal.

Supplement forms and doses

L-glutamine at 5 to 10 g daily for the gut-barrier side, split morning and evening on an empty stomach.

Collagen peptides at 10 to 15 g daily for the proline side, with vitamin C at 500 to 1,000 mg as a required cofactor rather than an optional extra.

Zinc carnosine at 75 mg twice daily targets the gut lining specifically. Adjust the relative doses toward whichever side of the ratio is driving the imbalance.
